Abstract
Chemical synthesis of long oligo deoxynucleotides via the catching-by-polymerization method has been used to address the various drawbacks encountered during traditional polymerase chain reaction assembly. However, the catching-by-polymerization method can encounter errors during synthesis. One of those errors, called substitution error, has been observed during the capping step of failure sequences after addition of deoxyguanosine nucleotides to the ODN strand. This report discusses a synthesis scheme during deoxyguanosine preparation to chlorinate the N2 carbon. This can prevent unintended nucleophilic attack during capping, which may result in the observed substitution error during final deprotection of synthesized oligo deoxynucleotides.
